data-sheets-value=\"{\"1\":2,\"2\":\"Galectinas y c\u00e1ncernnLas galectinas son prote\u00ednas que reconocen az\u00facares espec\u00edficos en las c\u00e9lulas y desencadenan o modulan de esta manera una respuesta biol\u00f3gica.nNuestro laboratorio fue pionero en demostrar que, a trav\u00e9s de su interacci\u00f3n con az\u00facares espec\u00edficos, Galectina-1 es capaz de inhibir la respuesta inmunol\u00f3gica y promover procesos que conducen a la progresi\u00f3n tumoral a las met\u00e1stasis.nDemostramos adem\u00e1s que las c\u00e9lulas tumorales utilizan Galectina-1 a los fines de inhibir la respuesta inmune antitumoral e inducir evasi\u00f3n del sistema inmunol\u00f3gico. En este sentido, Galectina-1 ha sido involucrada en el desarrollo de diversos tipos de c\u00e1ncer, y su expresi\u00f3n o sobreexpresi\u00f3n en tejidos tumorales es un signo de progresi\u00f3n maligna y de mal pron\u00f3stico .nNuestro equipo estudia estas interacciones entre prote\u00ednas y glicanos en diversos tipos tumorales como ser: C\u00e1ncer de mama, c\u00e1ncer de pulm\u00f3n, c\u00e1ncer renal, melanoma, c\u00e1ncer colorectal y leucemia linfoc\u00edtica cr\u00f3nica (LLC). nEl c\u00e1ncer de mama es una enfermedad muy frecuente, una de cada 8 mujeres va a padecerlo a lo largo de su vida. Si bien la detecci\u00f3n temprana y los avances en las terapias dirigidas hacen que la sobrevida de las pacientes sea realmente alta, a\u00fan quedan por descubrir nuevas estrategias para tratar la enfermedad metast\u00e1sica o diseminada. Nuestro equipo de trabajo de c\u00e1ncer de mama se dedica particularmente a estudiar c\u00f3mo las interacciones entre Galectina-1 y los az\u00facares que se encuentran sobre la c\u00e9lula tumoral pueden afectar la progresi\u00f3n tumoral y las met\u00e1stasis. Estudiamos principalmente el rol de Galectina-1, producida por el tumor o por el microambiente tumoral, desde dos aproximaciones fundamentales. Por un lado, evaluamos siGalectina-1 es capaz de frenar la respuesta inmunol\u00f3gica antitumoral al unirse a las c\u00e9lulas de nuestro sistema de defensas, y por otro analizamos si Galectina-1 est\u00e1 mediando ella misma la invasi\u00f3n y las met\u00e1stasis al actuar sobre las c\u00e9lulas del tumor.nEn particular en melanoma, en c\u00e1ncer de pulm\u00f3n y de ri\u00f1\u00f3n, estamos trabajando en descifrar los mecanismos que median la resistencia de estos tumores a las inmunoterapias m\u00e1s utilizadas como los anticuerpos bloqueantes de puntos de control inmunol\u00f3gico (anti PD-1 o anti PD-L1). Hemos establecido los modelos animales que reproducen la patolog\u00eda humana y nos permiten estudiar en detalle estos mecanismos.nnGalectina-1 y angiog\u00e9nesis tumoral nnNuestros trabajos sentaron las bases para explicar un nuevo mecanismo de resistencia a la terapia antiangiog\u00e9nica basado en la interacci\u00f3n entre Galectina-1 (Gal-1) y los glicanos presentes en la superficie de las c\u00e9lulas endoteliales que forman los vasos sangu\u00edneos. En 2014 publicamos un trabajo en la renombrada revista cient\u00edfica \u201cCell\u201d donde demostramos que los tumores resistentes a la terapia antiangiog\u00e9nica con Bevacizumab son capaces de modificar el perfil de glicanos de la superficie de las c\u00e9lulas endoteliales permitiendo el pegado de Gal-generando nuevos vasos sangu\u00edneos e ignorando el bloqueo de la terapia, inhibiendo as\u00ed, la efectividad del tratamiento anti angiogenico con anti-VEGF. Por otro lado, demostramos que Gal-1 tumoral no s\u00f3lonpromueve la resistencia a la terapia antiangiog\u00e9nica sino que adem\u00e1s, inhibe la respuesta inmunol\u00f3gica contra los tumores. Este efecto dual sabemos ahora que esnparte integral de un mecanismos de regulaci\u00f3n homeost\u00e1tica donde ambos sistemasninteraccionan mediante numerosos procesos celulares y moleculares. nnGalectinas y autoinmunidadnnNuestros hallazgos han demostrado que Galectin-1, al unirse a sus glicanos espec\u00edficos sobre la superficie celular, posee actividades antiinflamatorias y pro-apopt\u00f3ticas de los linfocitos T. Mientras que los linfocitos del tipo TH1- y TH-17 expresan el repertorio de glicanos de superficie que son cr\u00edticos para la uni\u00f3n de Gal-1 y la consecuente promoci\u00f3n de la apoptosis, los linfocitos TH2 est\u00e1n protegidos de la uni\u00f3n de Gal-1 a trav\u00e9s de la sialilaci\u00f3n diferencial de glicoprote\u00ednas de superficie. Estos mecanismos determinan que los ratones que son deficientes de Gal-1 desarrollan respuestas TH1- y TH17 aumentadas y presentan una mayor susceptibilidad a enfermedades autoinmunes como esclerosis m\u00faltiple, artritis reumatoidea, o uve\u00edtis. Nuestros hallazgos han identificado un nuevo v\u00ednculo molecular entre la glicosilaci\u00f3n diferencial de los linfocitos T helper, la susceptibilidad a la muerte celular y la inhibici\u00f3n de la respuesta inflamatoria. Este crucial descubrimiento puede servir para identificar un nuevo tratamiento en enfermedades autoinmunes cuya cura aun es una deuda con la sociedad. nn\"}\" data-sheets-userformat=\"{\"2\":14593,\"3\":{\"1\":0,\"3\":1},\"11\":4,\"14\":{\"1\":3,\"3\":1},\"15\":\"Arial\",\"16\":10}\">Galectins and cancer<\/span><\/p>\n<p>Galectins are proteins that recognize specific sugars in cells and thus trigger or modulate a biological response.<br \/>\nOur laboratory was a pioneer in showing that, through its interaction with specific sugars, Galectin-1 can inhibit the immune response and promoting processes that lead to tumor progression to metastasis.<br \/>\nWe further demonstrate that tumor cells use Galectin-1 for the purposes of inhibiting the antitumor immune response and inducing immune system evasion. In this sense, Galectin-1 has been involved in the development of various types of cancer, and its expression or overexpression in tumor tissues is a sign of malignant progression and poor prognosis.<br \/>\nOur team studies these interactions between galectins and glycans in various tumor types such as: breast cancer, lung cancer, kidney cancer, melanoma, colorectal cancer, and chronic lymphocytic leukemia (CLL).<br \/>\nBreast cancer is a very common disease, one in 8 women will suffer from it throughout her life. Although early detection and advances in targeted therapies make patient survival high, new strategies to treat metastatic or disseminated disease have yet to be discovered. Our breast cancer team is particularly dedicated to studying how the interactions between Galectin-1 and the sugars found on the tumor cell can affect tumor progression and metastasis. We mainly study the role of Galectin-1, produced by the tumor or by the tumor microenvironment, from two fundamental approaches. On the one hand, we evaluated whether Galectin-1 can stop the antitumor immune response by binding to the cells of our defense system, and on the other we analyzed whether Galectin-1 is itself mediating invasion and metastasis by acting on cells of the tumor.<br \/>\nIn particular, in melanoma, lung and kidney cancer, we are working on deciphering the mechanisms that mediate the resistance of these tumors to the most widely used immunotherapies such as immune checkpoint blockade antibodies (anti PD-1 or anti PD-L1). We have established animal models that reproduce human pathology and allow us to study these mechanisms in detail.<\/p>\n<p>Galectin-1 and tumor angiogenesis<\/p>\n<p>Our work set the bases to explain a new mechanism of resistance to antiangiogenic therapy based on the interaction between Galectin-1 (Gal-1) and glycans present on the surface of endothelial cells that form blood vessels. In 2014 we published a paper in the renowned scientific journal &#8220;&#8221;Cell&#8221;&#8221; where we demonstrated that tumors resistant to antiangiogenic therapy with Bevacizumab could modify the glycan profile on the surface their endothelial cells, allowing the adhesion of Gal-1 and generating new blood vessels. This resulted in the lack of response of antiangiogenic therapy, thus inhibiting the effectiveness of anti-VEGF treatment.
