potencial de las c\u00e9lulas NK en inmunooncolog\u00eda para catalizar la inmunidad tumoral<\/span><\/a><\/h4><\/div><div id=\"dbbf707bd8d49ed45\" class=\"panel-collapse collapse \" aria-labelledby=\"toggle_dbbf707bd8d49ed45\"><div class=\"panel-body toggle-content fusion-clearfix\">\n<p><strong>Descripci\u00f3n:<\/strong><\/p>\n<p>Las c\u00e9lulas citot\u00f3xicas naturales o c\u00e9lulas NK tienen la capacidad de eliminar c\u00e9lulas tumorales, producen citoquinas como el IFN-gamma y sesgan la respuesta inmune hacia un perfil Th1\/proinflamatorio y citot\u00f3xico a trav\u00e9s de su interacci\u00f3n con c\u00e9lulas dendr\u00edticas y macr\u00f3fagos. La comprensi\u00f3n de su rol en la eliminaci\u00f3n de c\u00e9lulas tumorales las ha posicionado a la vanguardia de la inmunooncolog\u00eda con el fin de desarrollar estrategias de inmunoterapia transformadoras para pacientes con c\u00e1ncer. Nuestros proyectos biotecnol\u00f3gicos tienen como objetivos: 1) identificar y validar de nuevos blancos moleculares de las c\u00e9lulas NK en inmunooncolog\u00eda y 2) capitalizar sus funciones efectoras mediante el empleo de anticuerpos monoclonales o sus derivados, con el fin de desarrollar modalidades de tratamiento de pacientes con c\u00e1ncer innovadoras. Estos anticuerpos monoclonales est\u00e1n dirigidos contra el Receptor de Transferrina, que es la v\u00eda de entrada de los virus Jun\u00edn, Machupo, Chapare, Guanarito y Sabi\u00e1, por lo que bloquean la internalizaci\u00f3n de part\u00edculas pseudotipadas de estos virus. Junto con colaboradores de los EEUU estamos realizando estudios in vitro e in vivo que demuestran la eficacia de estos anticuerpos, por lo que constituir\u00edan una potencial opci\u00f3n terap\u00e9utica para el tratamiento de esta familia de graves enfermedades infecciosas desatendidas de inter\u00e9s regional.<\/p>\n<p>En colaboraci\u00f3n con la empresa nacional Agropharma Salud Animal S.A. y a trav\u00e9s de varios convenios con el CONICET, estamos trabajando en el desarrollo de nuevas herramientas para diagnosticar la tuberculosis bovina. Uno de nuestros enfoques consiste en dise\u00f1ar prote\u00ednas recombinantes que, al ser aplicadas en la piel del animal, permiten detectar la infecci\u00f3n a partir de la respuesta del sistema inmunol\u00f3gico, similar a una prueba de alergia. Este m\u00e9todo se basa en un mecanismo natural del organismo llamado hipersensibilidad tipo IV.<\/p>\n<p>Adem\u00e1s, estamos desarrollando un an\u00e1lisis de laboratorio, conocido como ELISA, que permite identificar la presencia de anticuerpos contra la bacteria Mycobacterium bovis en animales infectados. A nivel celular, esto significa que los fibroblastos han envejecido y est\u00e1n perdiendo la capacidad para proliferar, migrar y sintetizar matriz extracelular. En este sentido, es de inter\u00e9s biom\u00e9dico el desarrollo y estudio de diferentes estrategias para revertir el fenotipo envejecido de fibroblastos d\u00e9rmicos y potenciar la capacidad de reparaci\u00f3n de piel. Esta modificaci\u00f3n tiene roles diversos, y suscit\u00f3 inter\u00e9s en el \u00e1mbito industrial al tiempo que diversas glicoprote\u00ednas (ej: anticuerpos monoclonales) comenzaron a utilizarse como bioterap\u00e9uticos.<\/p>\n<div>\n<p>En nuestro laboratorio ofrecemos una plataforma para el an\u00e1lisis estructural de glicanos (Glic\u00f3mica) por cromatograf\u00eda liquida. La caracterizaci\u00f3n estructural de glicanos se realiza por liberaci\u00f3n enzim\u00e1tica y\/o qu\u00edmica de los mismos, y su posterior marcaci\u00f3n fluorescente. Los glicanos as\u00ed tratados son analizados por cromatograf\u00eda l\u00edquida de intercambio ani\u00f3nico (WAX), con el objeto de establecer el porcentaje de glicanos cargados negativamente. Para un an\u00e1lisis estructural completo, la muestra se analiza por cromatograf\u00eda l\u00edquida de interacci\u00f3n hidrof\u00edlica (HPLC\/UPLC-HILIC), una de las t\u00e9cnicas m\u00e1s sensibles para la caracterizaci\u00f3n estructural de posibles cambios en los perfiles de glicosilaci\u00f3n. Se detectan glicanos en el orden de femtomoles, siendo adem\u00e1s cuantitativa ya que la se\u00f1al es directamente proporcional a la aglicona fluorescente.
