#3366ff;\"><strong><u>Biomarcadores de respuesta a la terapia del c\u00e1ncer basados en prote\u00ednas quinasas.<\/u><\/strong><\/span><\/p>\n<p>Directora: Dra. Virginia Novaro.<\/p>\n<p>La perspectiva de nuestro laboratorio es avanzar en el conocimiento de los mecanismos mediados por las v\u00edas de PI3K\/AKT\/mTOR y ciclina D1\/CDK4\/6\/Rb en la progresi\u00f3n del c\u00e1ncer mamario de tipo luminal y eventualmente, definir nuevos marcadores asociados con la evoluci\u00f3n de la enfermedad. El impacto de nuestra investigaci\u00f3n reside en que la activaci\u00f3n de dichas v\u00edas de se\u00f1alizaci\u00f3n se propone como v\u00edas de escape tumoral fundamentalmente durante la resistencia a las terapias sist\u00e9micas convencionales. En los \u00faltimos 5 a\u00f1os se han incorporado a la cl\u00ednica nuevos inhibidores espec\u00edficos contra ellas, tales como inhibidores de PI3K, de mTOR y de CDK4\/6. Sin embargo, a\u00fan no hay consenso sobre cu\u00e1les son los marcadores moleculares de activaci\u00f3n de cada v\u00eda que mejor se asocian con la evoluci\u00f3n cl\u00ednica de las pacientes, as\u00ed como tampoco se conoce c\u00f3mo se modifica la activaci\u00f3n de dichas v\u00edas luego de exponer a las pacientes a las terapias dirigidas. Por ello, queremos profundizar el conocimiento sobre el rol de PI3K\/AKT\/mTOR y ciclina D1\/CDK4\/6\/Rb en los mecanismos implicados en la progresi\u00f3n de la enfermedad y en la respuesta del c\u00e1ncer de mama a los agentes terap\u00e9uticos disponibles. Para ello, evaluamos tumores de pacientes que cursan la enfermedad para encontrar marcadores moleculares, y utilizamos modelos experimentales de tumores de mama inducidos en ratones, as\u00ed como xenoinjertos de l\u00edneas celulares tumorales, para ensayar las opciones terap\u00e9uticas m\u00e1s beneficiosas seg\u00fan los marcadores encontrados.<\/p>\n<p>Las l\u00edneas de trabajo que desarrollamos son:<\/p>\n<p>1) Uso de las isoformas AKT1 y AKT2 como factores pron\u00f3stico en c\u00e1ncer de mama;<\/p>\n<p>2) Estudio de la interacci\u00f3n entre las v\u00edas PI3K\/AKT\/mTOR y CDK4-6\/Rb\/ciclina D1 como factor predictivo de respuesta al tratamiento anti-tumoral;<\/p>\n<p>3) Determinaci\u00f3n de microRNAs espec\u00edficos involucrados en la regulaci\u00f3n diferencial de prote\u00ednas quinasas para disponer de marcadores perif\u00e9ricos de diagn\u00f3stico y\/o pron\u00f3stico.<\/p>\n<p><span style=\"color: #3366ff;\"><strong><u>Biomarcadores para el diagn\u00f3stico temprano del c\u00e1ncer de mama basados en miRNAs circulantes. <\/u><\/strong><\/span><\/p>\n<p>Directora: Dra. Adriana De Siervi.<\/p>\n<p>Los miRNAs son mol\u00e9culas promisorias para ser utilizadas como biomarcadores ya que tienen la capacidad de ser liberadas por un tumor (aun cuando este es indetectable) y que pueden circular en la sangre de los pacientes de manera precoz e incluso de manera diferencial con los distintos tratamientos. Es por eso que se los puede utilizar en \u201cbiopsias l\u00edquidas\u201d, es decir detectar los miRNAs en la sangre u otros fluidos corporales para el diagn\u00f3stico de enfermedades. Esta es una herramienta muy esperanzadora para el diagn\u00f3stico, pron\u00f3stico y seguimiento de los pacientes, ya que es un m\u00e9todo no invasivo para el paciente.<\/p>\n<p>Por otro lado, la neoadyuvancia es el tratamiento quimioter\u00e1pico sist\u00e9mico que se realiza previamente al tratamiento quir\u00fargico del c\u00e1ncer en general. La neoadyuvancia ha permitido reestadificar a los pacientes, disminuir la progresi\u00f3n tumoral e incluso valorar la respuesta al tratamiento <em>in vivo<\/em>. Considerando que el uso de las biopsias l\u00edquidas basadas en miRNAs para el seguimiento y pron\u00f3stico de pacientes con c\u00e1ncer de mama en neoadyuvancia a\u00fan no se ha establecido, actualmente estamos investigando los miRNAs circulantes en pacientes con c\u00e1ncer de mama previo y post-neadyuvancia para generar nuevos biomarcadores para el pron\u00f3stico del c\u00e1ncer de mama y definir la estrategia terap\u00e9utica espec\u00edfica. Asimismo, estudios precl\u00ednicos de resistencia al tratamiento neadyuvante permitir\u00e1n caracterizar miRNAs que puedan utilizarse para mejorar la respuesta terap\u00e9utica en pacientes con c\u00e1ncer de mama.<\/p>\n<p><span style=\"color: #3366ff;\"><strong><u>Biomarcadores para la terapia del c\u00e1ncer de mama triple negativo basados en miRNAs. <\/u><\/strong><\/span><\/p>\n<p>Directora Dra. Paola De Luca.<\/p>\n<p>Nuestro laboratorio estudia hace a\u00f1os el rol de los miRNAs en el desarrollo y progresi\u00f3n del c\u00e1ncer de mama, as\u00ed como su potencial en el diagn\u00f3stico, pron\u00f3stico y terapia de esta enfermedad.\u00a0 El subtipo molecular triple negativo es el de peor pron\u00f3stico y con menores opciones terap\u00e9uticas. Adem\u00e1s, se sabe que los factores no heredables tienen un alto impacto en el riesgo y progresi\u00f3n del c\u00e1ncer. El s\u00edndrome metab\u00f3lico es un factor de riesgo para el c\u00e1ncer de mama que aumenta su agresividad y met\u00e1stasis, y presenta mayor prevalencia en el c\u00e1ncer de mama triple negativo. El objetivo de esta l\u00ednea de investigaci\u00f3n es generar terapias dirigidas para el c\u00e1ncer de mama triple negativo asociado o no al s\u00edndrome metab\u00f3lico basadas en miRNAs. Para esto, llevamos a cabo microarreglos de expresi\u00f3n y an\u00e1lisis bioinform\u00e1ticos para identificar los miRNAs claves en el crecimiento y progresi\u00f3n del c\u00e1ncer de mama triple negativo, asociado o no al s\u00edndrome metab\u00f3lico.
