sintetico Tibolona revierte la reacci\u00f3n glial, la hiperexpresi\u00f3n de factores proinflamatorios TLR4 y HMGB1 y la activaci\u00f3n del eje NLRP3-caspasa1-IL-1B. Se atenua la v\u00eda Akt\/NKkB y limita la desmielinizaci\u00f3n. Mejoran s\u00edntomas neurol\u00f3gicos sin compromiso \u00faterino y representa una terap\u00e9utica prometedora para la inflamaci\u00f3n en la EM. Considerando que los modelos de EM presentan hiperactivaci\u00f3n HPA, una l\u00ednea futura investigara los efectos de antimineralocorticoides (Eplerenona) y antiglucocorticoides (CORT113176) sobre aspectos moleculares y cl\u00ednicos de EM. Hip\u00f3tesis: antagonizando estos receptores se bloquear\u00edan par\u00e1metros neuroinflamatorios aparejando la mejor\u00eda cl\u00ednica. \\n\\nNeuroprotecci\u00f3n en un modelo de esclerosis lateral amiotrofica (ELA): ELA es una enfermedad devastadora con muerte por falla respiratoria y degeneraci\u00f3n de motoneuronas. La incidencia es 2,5:1 (hombre:mujer); sin embargo, en mujeres posmenop\u00e1usicas \u226570 a\u00f1os la relaci\u00f3n hombre:mujer se iguala, indicando a rol de las hormonas sexuales. El tratamiento con allopregnanolona o testosterona en un modelo de ELA, el rat\u00f3n Wobbler, reduce astro y microgliosis, el estr\u00e9s oxidativo y enlentece la enfermedad. Postulamos que el efecto neuroprotector depende de la modulaci\u00f3n de la maquinaria esteroidog\u00e9nica. A futuro, analizaremos la expresi\u00f3n de enzimas esteroidog\u00e9nicas en SNC y gl\u00e1ndulas end\u00f3crinas. Hip\u00f3tesis: la regulaci\u00f3n de la neuroesteroidog\u00e9nesis y de mol\u00e9culas en la membrana de asociaci\u00f3n mitocondria-ret\u00edculo endopl\u00e1smico (MAM) interviene en la degeneraci\u00f3n de motoneuronas. Postulamos que el receptor sigma1, que une esteroides sexuales, regularia de la neuroesteroidog\u00e9nesis.\\n\\nEfectos proinflamatorios de los glucocorticoides (GC): Los pacientes con ELA y el rat\u00f3n Wobbler presentan hipercorticoidismo e hiperfuncion HPA. Actualmente estudiamos si el bloqueo de los receptores para GC (GR) previene la neuroinflamacion espinal. Los moduladores del GR CORT113176 y CORT 108297 ofrecen neuroprotecci\u00f3n y disminuyen los mediadores TNF\u03b1, HMGB1 (alarmina, high mobility group box 1 protein), TLR4, MyD95 y NFkB, que aumentan la transcripci\u00f3n de citoquinas inflamatorias. Como linea futura emplearemos controles con neuroinflamacion asociada al estr\u00e9s cr\u00f3nico, proceso mediado por GC y que proponemos inhibir por antagonistas GR. Hipotesis: El estr\u00e9s cr\u00f3nico conduce a la inflamaci\u00f3n (la espada de doble filo) y si los cambios moleculares, metabolicos y comportamentales son susceptibles de tratamiento farmacol\u00f3gico.\\n \\nGenisteina y s\u00edndrome metabolico (SM) en la rata espontanemente hipertensa : El SM es un factor de riesgo cardiovascular. Desarrollamos SM alimentando ratas SHR con dieta alta en grasas y carbohidratos, las que muestran neuroinflamacion, baja neurogenesis y d\u00e9ficits cognitivos. Estudiamos al fitoestrogeno geniste\u00edna (GEN) que se une al ER\u03b2 y al GPER y no posee efectos feminizantes en machos. La GEN tiene efectos neuroprotectores, anti-inflamatorios y mejora el comportamiento. Hip\u00f3tesis: la Genisteina posee efectos neuroprotectores y antiinflamatorios y por lo tanto mejorar\u00eda la neuropatolog\u00eda y el d\u00e9ficit cognitivo del SM. Tambi\u00e9n estudiaremos su mecanismo de acci\u00f3n utilizando agonistas\/antagonistas espec\u00edficos de ER\u03b1 y ER\u03b2.\\n\\nLesi\u00f3n de la medula espinal y alteraciones encef\u00e1licas. Describimos en la lesi\u00f3n espinal (LME) cambios hipocampales y deterioro cognitivo, debidos a la neuroinflamaci\u00f3n hipocampal cr\u00f3nica acompa\u00f1ada de activaci\u00f3n de microglia y astrocitos y producci\u00f3n de citoquinas pro-inflamatorias. Esto se acompa\u00f1a de disminuci\u00f3n de neurogenesis y neurodegeneraci\u00f3n hipocampal, explicando el deterioro cognitivo. En la actualidad estudiamos el rol de los GC en la pre-activaci\u00f3n de c\u00e9lulas gliales y en la neuroinflamaci\u00f3n cr\u00f3nica. Hip\u00f3tesis: la acci\u00f3n de los GC en la etapa aguda determinar\u00eda la pre-activaci\u00f3n microglial con activacion en la etapa cr\u00f3nica por degeneraci\u00f3n transneuronal. Considerando que los modelos de EM presentan hiperactivaci\u00f3n HPA, una l\u00ednea futura investigar\u00e1 los efectos de antimineralocorticoides (Eplerenona) y antiglucocorticoides (CORT113176) sobre aspectos moleculares y cl\u00ednicos de EM. La incidencia es 2,5:1 (hombre:mujer); sin embargo, en mujeres posmenop\u00e1usicas \u226570 a\u00f1os la relaci\u00f3n hombre:mujer se iguala, indicando a rol de las hormonas sexuales. El tratamiento con allopregnanolona o testosterona en un modelo de ELA, el rat\u00f3n Wobbler, reduce astro y microgliosis, el estr\u00e9s oxidativo y enlentece la enfermedad. Postulamos que el efecto neuroprotector depende de la modulaci\u00f3n de la maquinaria esteroidog\u00e9nica. A futuro, analizaremos la expresi\u00f3n de enzimas esteroidog\u00e9nicas en SNC y gl\u00e1ndulas end\u00f3crinas. Hip\u00f3tesis: la regulaci\u00f3n de la neuroesteroidog\u00e9nesis y de mol\u00e9culas en la membrana de asociaci\u00f3n mitocondria-ret\u00edculo endopl\u00e1smico (MAM) interviene en la degeneraci\u00f3n de motoneuronas. Postulamos que el receptor sigma1, que une esteroides sexuales, regularia de la neuroesteroidog\u00e9nesis.<\/p>\n<p><strong>Efectos proinflamatorios de los glucocorticoides<\/strong> (GC): Los pacientes con ELA y el rat\u00f3n Wobbler presentan hipercorticoidismo e hiperfuncion HPA. Actualmente estudiamos si el bloqueo de los receptores para GC (GR) previene la neuroinflamaci\u00f3n espinal. Los moduladores del GR CORT113176 y CORT 108297 ofrecen neuroprotecci\u00f3n y disminuyen los mediadores TNF\u03b1, HMGB1 (alarmina, high mobility group box 1 protein), TLR4, MyD95 y NFkB, que aumentan la transcripci\u00f3n de citoquinas inflamatorias. Como linea futura emplearemos controles con neuroinflamaci\u00f3n asociada al estr\u00e9s cr\u00f3nico, proceso mediado por GC y que proponemos inhibir por antagonistas GR. Desarrollamos SM alimentando ratas SHR con dieta alta en grasas y carbohidratos, las que muestran neuroinflamacion, baja neurogenesis y d\u00e9ficits cognitivos. Estudiamos al fitoestrogeno geniste\u00edna (GEN) que se une al ER\u03b2 y al GPER y no posee efectos feminizantes en machos. La GEN tiene efectos neuroprotectores, anti-inflamatorios y mejora el comportamiento. Hip\u00f3tesis: la Genisteina posee efectos neuroprotectores y antiinflamatorios y por lo tanto mejorar\u00eda la neuropatolog\u00eda y el d\u00e9ficit cognitivo del SM. Describimos en la lesi\u00f3n espinal (LME) cambios hipocampales y deterioro cognitivo, debidos a la neuroinflamaci\u00f3n hipocampal cr\u00f3nica acompa\u00f1ada de activaci\u00f3n de microgl\u00eda y astrocitos y producci\u00f3n de citoquinas pro-inflamatorias. Esto se acompa\u00f1a de disminuci\u00f3n de neurog\u00e9nesis y neurodegeneraci\u00f3n hipocampal, explicando el deterioro cognitivo. En la actualidad estudiamos el rol de los GC en la pre-activaci\u00f3n de c\u00e9lulas gliales y en la neuroinflamaci\u00f3n cr\u00f3nica. Hip\u00f3tesis: la acci\u00f3n de los GC en la etapa aguda determinar\u00eda la pre-activaci\u00f3n microglial con activaci\u00f3n en la etapa cr\u00f3nica por degeneraci\u00f3n transneuronal.
